5-Amino-1MQ (5-amino-1-methylquinolinium) is a small-molecule, membrane-permeable inhibitor of nicotinamide N-methyltransferase (NNMT), the cytosolic enzyme that methylates nicotinamide to 1-methylnicotinamide using S-adenosylmethionine.
Neelakantan and colleagues characterized the methylquinolinium series in Caco-2 permeability assays and cultured 3T3-L1 adipocytes, reporting selectivity over related SAM-dependent methyltransferases and NAD+-salvage enzymes, reduced intracellular 1-methylnicotinamide, and increased NAD+ and SAM, in a 2017 Biochemical Pharmacology paper on selective small-molecule NNMT inhibitors. It is used in NNMT enzymology and NAD+/SAM flux studies.
