FOXO4-DRI is a D-retro-inverso peptide built from the FOXO4 sequence that binds p53; the D-amino-acid, reversed-sequence design gives it protease resistance while preserving the interaction surface. It is used as a tool compound to disrupt the FOXO4–p53 interaction in senescent cells.
Kong and colleagues applied FOXO4-DRI to keloid-derived senescent fibroblasts and organ cultures and reported apoptosis accompanied by nuclear exclusion of serine-15-phosphorylated p53, in a 2025 Communications Biology paper on FOXO4-DRI in senescent fibroblasts.
