PT-141 (bremelanotide) is the cyclic heptapeptide Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, the C-terminal carboxylic-acid analog of Melanotan II. It is a non-selective agonist at the melanocortin receptors MC1R, MC3R, MC4R and MC5R.
The receptor pharmacology of this cyclic-lactam α-MSH(4-10) scaffold was mapped by Hruby and colleagues at cloned human and mouse melanocortin receptors, in a 1995 Journal of Medicinal Chemistry paper on cyclic lactam α-melanotropin analogs. PT-141 is used in melanocortin-receptor binding, cAMP, and receptor-selectivity assays.
