SS-31 (elamipretide, MTP-131) is the Szeto-Schiller tetrapeptide D-Arg-Dmt-Lys-Phe-NH2, an aromatic-cationic peptide that partitions into the inner mitochondrial membrane, where the anionic phospholipid cardiolipin is enriched.
Mitchell and colleagues analyzed its interaction with model and mitochondrial membranes using biophysical and computational approaches, reporting that the peptide binds the membrane interfacial region in proportion to surface charge and modulates surface electrostatics without destabilizing the bilayer, in a 2020 Journal of Biological Chemistry paper on how SS-31 binds lipid bilayers. It is used in isolated-mitochondria, liposome, and membrane-biophysics studies.
