Ipamorelin is a synthetic pentapeptide, Aib-His-D-2-Nal-D-Phe-Lys-NH2, developed at Novo Nordisk as a growth-hormone secretagogue-receptor (GHS-R1a, the ghrelin receptor) agonist. It lacks the central Ala-Trp dipeptide of GHRP-1 and was identified from a structure-activity series built around that deletion.
In the original characterization, Raun and colleagues reported that ipamorelin released growth hormone from primary rat pituitary cells with potency comparable to GHRP-6 and, unlike GHRP-2 and GHRP-6, did not stimulate ACTH or cortisol in the animal models tested, which is why the paper describes it as the first selective growth hormone secretagogue. It is used in receptor-selectivity, pituitary cell-culture, and GHS-R1a signaling studies.
